Choline has several essential biological roles

The body uses choline to make phosphatidylcholine and sphingomyelin for cell membranes, acetylcholine for nervous-system signaling, and methyl-group metabolites involved in metabolism. The liver makes some choline, but not enough to meet all human needs.

Potential effects and evidence map

Evidence has two distinct layers. Choline essentiality, membrane biology, acetylcholine synthesis, methyl-group metabolism, and U.S. intake guidance are well established. Nootropic efficacy is much less certain and depends on the exact form and population. Acute choline bitartrate studies found no memory improvement about 60 minutes after a large single dose. A 2024 crossover study of only 20 resistance-trained men reported Alpha-GPC Stroop improvements at 60 minutes, but the author advised the sponsor and the trial was retrospectively registered. A manufacturer-sponsored 12-week citicoline trial reported episodic-memory benefits in older adults with age-associated memory impairment, yet EFSA concluded in 2024 that the total evidence did not establish a cause-and-effect relationship for memory. A separate 12-week Alpha-GPC trial reported benefit in people diagnosed with mild cognitive impairment; it does not validate plain choline or routine healthy-user enhancement.
